ANALYTICAL METHOD DEVELOPMENT AND VALIDATION OF GLIPIZIDE IN BULK AND TABLET DOSAGE FORM BY UV-SPECTROPHOTOMETRIC METHOD (FIRST-ORDER DERIVATIVE)

ABSTRACT
A new, simple, accurate, and precise first-order derivative spectroscopic method was developed and validated to estimate Glipizide in bulk and Tablet dosage form. The stock solution was prepared by weighing 100 mg of standard Glipizide in a 100 ml volumetric flask with 0.1N KOH. The stock solution was made to produce 1000 µg/ml with 0.1N KOH. Further dilutions were prepared as per the procedure. The drug solution showed the maximum absorbance at 275 nm. The linearity was found in the concentration range of 6-30 µg/ml. The correlation coefficient was found to be 0.998. The regression equation was found to be Y=0.029x+0.009. The method was validated for linearity, accuracy, precision, ruggedness, robustness, LOD, and LOQ. The LOD and LOQ for estimation of Glipizide were found to be 0.351 µg/ml and 1.064 µg/ml respectively. Recovery of Glipizide was found to be in the range of 102-112 %. The proposed method was successfully applied for the quantitative determination of Glipizide in bulk and Tablet dosage forms.
