ABSTRACT
In situ gelling formulations are drug delivery system which typically exist in a liquid form at room temperature and change into gel state after application to the body in response to various stimuli such as change in temperature, ph and ionic composition. In situ gelling drug delivery systems have gained enormous attention over the last decade. Such systems can be administered through different routes, to achieve local or systematic drug delivery and can also be successfully used as vehicles for drug-loaded nano-particles and microparticles. This transformation allows for localized and sustained drug release, reducing the frequency of administration and minimizing systemic side effects. In situ gels are widely explored across various routes of administration, including ocular, nasal, oral, injectable, vaginal, and transdermal delivery, showcasing their adaptability and effectiveness. The incorporation of biocompatible polymers like poloxamers, chitosan, and gellan gum enables the formulation of responsive systems tailored to specific therapeutic needs. Applications range from targeted drug delivery for chronic diseases to localized treatment of infections and cancer. This abstract highlights the potential of in situ gels to revolutionize drug delivery by combining ease of administration, prolonged drug residence, and enhanced bioavailability.
