FORMULATION AND EVALUATION OF TRANSDERMAL PATCH OF PIOGLITAZONE HCl

ABSTRACT
The purpose of this research was to develop a transdermal patch of pioglitazone HCl using the polymer HPMC K15M, Eudragit RS100 by the solvent evaporation technique by using 20% w/v of PEG 400 to the polymer weight as plasticizer and 15% w/v of DMSO as permeation enhancer. The physicochemical compatibility of the drug and the polymer studies by infrared spectroscopy. Formulated transdermal patches were physically evaluated with regard to thickness, weight variation, folding endurance, flatness, % Moisture uptake, % Moisture loss, % Drug content. Invitro permeation studies of formulations were performed by using 3 stage Franz diffusion cells. Variations in drug permeation profile were observed among various formulations, from the all formulations, Formulation PH-2 was selected as the best formulation by using the 2factor central composite design. Prepared PH-2 selected central composite formulation indicated good physical stability. It shown the drug release follows zero order and the mechanism of release is diffusion from the polymer.
